The Adenosine Deaminase (ADA) Test Kit is a clinical specific protein enzymatic assay designed for the quantitative determination of Adenosine Deaminase (ADA) activity in human serum or plasma. ADA is a highly sensitive biomarker reflecting hepatocellular injury. It serves as an essential routine liver function test to evaluate acute liver injury, detect residual diseases, assist in the diagnosis of chronic liver diseases (such as chronic hepatitis), and contribute to the prognostic evaluation of liver fibrosis.
The assay operates on a coupled multi-enzymatic reaction. First, ADA present in the sample enzymatically degrades adenosine to produce inosine. Inosine is then converted to hypoxanthine through the action of purine nucleoside phosphorylase (PNP). Subsequently, hypoxanthine is oxidized by xanthine oxidase (XOD) to yield uric acid and hydrogen peroxide (H₂O₂). Under peroxidase (POD) action, H₂O₂ reacts with EHSPT and 4-aminoantipyrine (4-AAP) to form colored quinone dyes. A specific protein analyzer measures the rate of increase in absorbance of these quinone complexes (which is directly proportional to ADA activity) and calculates the final concentration against a standard calibration curve.
